The Effect of Aspirin on Platelet Aggregation

Abstract

|
To study the effect of Aspirin on the bleeding tendency, we observed the platelet aggregation in 14 normal healthy persons before and after the Aspirin administration with platelet aggregometer. Eleven persons took single dose of Aspirin ranging from 1.4mg/kg of body weight to 32mg/kg and three persons took Aspirin ranging from 1.9 mg/kg/day to 19 mg/kg/day for 4-5 days. Platelet aggregation was observed before and at 30 min. 90 min. 3 days, 4 days, 5 days, 6 days and 7 days after the administration of Aspirin. After Aspirin ingestion the primary platelet aggregation curve was well preserves but he secondary aggregation curve by adenosine-5¢¥-diphosphate (1.2¥ìM-2.4,¥ìM) and epinephrine (8¥ìg) disappeared.
The degree of aggregation by collagen was also markedly decreased with Aspirin. The minimal effective dose of Aspirin was 2.0mg/kg of body weight and platelet dysfunction appeared since 30 min. after Aspirin ingestion and persisted for 3-6 days. The degree and duration of platelet dysfunction were not related to the dose of Aspirin.
